The owner of the shop could be her or a bike enthusiast like Jay Leno, or a business person who doesn't know squat about motorbikes (an interesting twist). Guest appearances by other bike enthusiast who interact with Flo, to shop for accessories or bikes (males and/or females). She's a strong female, not married, but has a circle of biker friends. She doesn't have children, but sometimes tends to her 8 - 10 yr old nephew. She has siblings (sister played by Lauren Graham, that would be too cool!) How 'bout the location - in a small town - like Blowing Rock or Boone, North Carolina. Lots of bikers travel on the Blue Ridge Parkway. Many stories could be told of their adventures.
